Cell-free transcription-translation (TXTL) systems can be used for rapid and flexible protein expression and screening workflows, powering protein engineering and discovery, synthetic biology, AI-driven approaches, biomanufacturing, and more. However, reliance on plasmid DNA templates for expression requires time-consuming cloning steps, limiting speed and efficiency.

In this webinar, supported by Elegen and hosted by DDW, Dr Galit Meshulam-Simon, PhD from Elegen, and Dr Ben Steil, PhD from Daicel Arbor Biosciences will discuss how combining Elegen’s ability to synthesise high-quality linear DNA up to 7kb and successful expression in cell-free systems such as myTXTL® Pro from Daicel Arbor Biosciences, enable rapid, efficient and streamlined in vitro workflows.

It will be demonstrated that ENFINIA™ Linear DNA templates are functional in myTXTL® Pro and drive reporter gene expression at levels comparable to plasmid DNA templates. Additionally, a case study for a rapid CRISPR-Cas activity assay will be presented. In this study, successful co-expression of a Cascade operon expression construct and gRNA in a TXTL reaction accelerated the design-build-test-learn cycle to eight days, bypassing traditional cloning and purification steps required by conventional workflows.

A Q&A session follows the webinar presentations.
